Metabolite Profiling - With Untargeted and Targeted Platforms

Metanomics Health utilizes two highly complementary mass spectrometry technologies – LC-MS/MS plus GC-MS – in parallel.

This combination allows the analysis of the widest possible chemical range of metabolites. Continuous operation of robotics-based sample extraction and processing and more than 75 mass spectrometers in a largely automated high-throughput facility provide the capacity for 400,000 analyses per year.

A modular set-up still allows full flexibility for customized solutions. Unique in the industry, Metanomics Health employs two analytical approaches in parallel. A wide range of pre-specified and identified metabolites are analyzed on so-called target platforms. These metabolites of direct commercial or scientific importance provide immediate insight into the specific performance of an organism. In parallel, and different from conventional approaches, metanomics applies broad metabolite profiling (MxP™ Broad Profiling) to monitor an exceptional range of additional compounds, irrespective of whether they are known or currently unknown. This latter approach has proven to deliver discoveries of “unexpected”, novel metabolic markers and compounds.
Novel and proprietary software systems ensure precise result validation and compound annotation, even for unknown analytes.
Metabolite biomarkers provide maximum value, commercial and scientific, only as annotated biochemical compounds. metanomics’ bioanalytical platform provides this information and capability.
Metanomics Metabolite Profiling

Untargeted Metabolite Profiling: MxP™ Broad Profiling

Samples are extracted by a proprietary method yielding a lipid and a polar fraction each of which are used for GC-MS and LC-MS/MS analyses (4-Phase- Profiling). The metabolites are analyzed in a semi-quantitative approach. Metabolite classes typically analyzed by this method include:

  • Amino acids and related
  • Carbohydrates
  • Energy metabolites
  • Nucleobases
  • Vitamins and co-factors
  • Miscellaneous, e.g. creatine, creatinine
  • Lipids

    • Fatty acids (free fatty acids and those derived from higher lipids)
    • Cholesterol
    • Bile acids
    • Fatty alcohols and related
    • DAGs, TAGs

Typically between 200 and 500 metabolites can be analyzed above level of quantification in common biological matrices.
The broad profiling platform is tailored to detect metabolite changes occurring in a large variety of applications related to pharmacology, nutrition, diagnostics and toxicology.

Hormone Platform (MxP™ Catecholamines and MxP™ Steroids)

A targeted panel of low abundance catecholamines and derivatives and a panel of steroid hormones are analyzed by a specific Solid Phase Extraction(SPE)-LC-MS/MS method. These metabolites are analyzed with absolute quantitation or relative quantitation.

The hormone platform adds additional value, e.g. in the areas of neurological disorders, hormonal regulation of metabolism, reproductive toxicology and certain physiological conditions such as stress or pain

Lipidomics: MxP™ Lipids

Lipid extracts are fractionated into eleven different lipid groups ranging from neutral lipids to phospholipids.

The fractions are analyzed by GC-MS after derivatization yielding the fatty acid methyl esters (FAME) corresponding to the acyl moieties of the class-separated lipids.

The fractions are further analyzed by LC-MS/MS for of cholesterol esters and sphingoymelins, respectively.
Lipid profiling offers additional insights into cellular processes affected by e.g. adipositas, diabetes, cardiovascular diseases and many other diseases.

MxP™ Energy

Method applicable to cellular sample types. Phosphorylated or carboxylated metabolites like ATP, NADH, intermediates of glycolysis, mevalonate pathway, purine and pyrimidine metabolism, pentose phosphate cycle and Krebs cycle, are extracted and analyzed semiquantitatively with a special UPLC-MS/MS method.
The energy metabolite platform is used to describe physiological conditions affecting the energy status of a cell as well as cell proliferation or apoptosis, processes of high relevance in the area of oncology

MxP™ Eicosanoids

A panel of eicosanoic acid derivatives are analyzed by a specific SPE-LC-MS/MS method. These metabolites are analyzed with absolute quantitation.
Eicosanoids yield valuable information, e.g. when studying diseases with an inflammatory component such as cancer, atherosclerosis, cardiovascular disease or immunological diseases.
